A critical aspect of our experimental plans is to develop methods that enable us to properly phenotype the mutant animal models we generate. Within the Division there is a particular strength in the development and use of 3D imaging techniques. We have been using these to analyse our experimental animal models and to produce a 3D anatomy database. We have developed protocols using Optical Projection Tomography (OPT) that enabled us to describe to a previously unobtainable level of resolution, the limb-type specific characteristics that were transformed by the activity of Pitx1. We also used these 3D imaging methods to produce a Mouse 3D Limb Anatomy database. We have also used High resolution Episcopic microscopy (HREM) and OPT in a recent study to characterise with high resolution, limb soft tissue patterning defects.
