High throughput reverse genetics/TILLING

We are collaborating with the Stemple group at the Sanger Institute and the Harland lab at UC Berkeley to create a Xenopus Mutation Resource for use in reverse genetic TILLING strategies.  TILLING (Targeting Induced Local Lesions In Genomes) identifies carriers of mutations in specific sequences by amplifying and characterizing sequences from a large library of genomic DNAs harvested from individual mutagenized animals. Once a carrier of a mutation is identified, it can be recovered either from a cognate library of living animals or frozen sperm for phenotypic characterization.

Targetting Induced Local Lesions In Genomes is a reverse genetic method based on sequencing amplicons(?) from mutagenized genomes for mutations in specific genes. Genomes bearing mutations in the gene of interest are recovered from a matching library of frozen sperm or living frogs.
